I have had several Emails concerning the morphology of plaques produced by phages that produce high concentrations of polysaccharide depolymerase in infected hosts. I thought it might be useful to summarise these discussions.
If the host has capsules comprised of polysaccharides then many phages code for a polysaccharide depolymerase enzyme to assist with the infection processes. Such phages can generally be recognised by halo production-a secondary zone of lysis around the main plaque. Regretfully I do not have any photographs of the plaques produced by polysaccharide depolymerase producing phages for lactic acid bacteria. Readers interested in this subject might find the image below useful. This shows the morphology of plaques formed by a phage for a azotobacter that produces a capsule-digesting enzyme.
